Title: Tracing the Dynamics of Stem Cell Fate Directed by Niches
BIOGRAPHY: Joo-Hyeon Lee is a Wellcome Senior Fellow and a Group Leader at the Wellcome – MRC Cambridge Stem Cell Institute, University of Cambridge. Her group focuses on understanding how stem and progenitor cells regulate their fate behaviours to specify and maintain tissues. They ask fundamental questions involving how stem cells sense environmental changes and determine their cell fate, and how niches develop and remodel the local environment during lung regeneration and the early stages of disease progression. Joo-Hyeon was awarded a Wellcome Sir Henry Dale Fellowship, a ERC starting grant, and a Suh Kyungbae Foundation Young Investigator award.
ABSTRACT: While the acquisition of cellular plasticity in adult stem cells is essential for rapid regeneration after tissue injury, the precise cellular events and molecular mechanisms that direct stem cell fate behaviours remain elusive. I will present the dynamics of stem cell differentiation plasticity that are coordinated by their niches. By utilising a combination of in vivo genetic lineage tracing, single-cell profiling, quantitative clonal analysis and ex vivo organoid culture, we defined a step-wise trajectory of stem cell differentiation and key
regulatory networks at individual levels during lung regeneration after injury. I will also further discuss how the regenerative process can be hijacked into disease modelling via modulating stem cell fate dynamics in lung diseases.
